Botulinum toxin

Botulinum toxin (BoNT) is a 150-kDa bacterial neurotoxin produced by Clostridium botulinum, a gram-positive anaerobic bacterium. It is proteolytically processed in vivo into a biologically active di-chain form consisting of a heavy chain (~100 kDa) and a light chain (~50 kDa), linked by a single disulfide bond. Seven serotypes (A through G) are known, each with distinct SNARE-protein targets and duration profiles.

BoNT's mechanism is fundamentally different from other neuromuscular peptides in our catalog, including Snap-8. Where Snap-8 acts through competitive, reversible SNAP-25 binding, botulinum toxin is a zinc-dependent endopeptidase: its light chain irreversibly cleaves specific peptide bonds within SNARE complex proteins. BoNT-A and BoNT-C cleave SNAP-25 itself; BoNT-B, BoNT-D, BoNT-F, and BoNT-G cleave synaptobrevin (VAMP), a vesicle-docking protein. The third target, syntaxin, is cleaved by BoNT-C. Cleavage of any one of these proteins completely prevents the assembly of functional SNARE complexes, blocking acetylcholine exocytosis irreversibly until the neuron synthesizes new protein, a process taking weeks to months. This distinguishes BoNT as a long-acting, near-complete neuromuscular blockade tool, in contrast to Snap-8's reversible partial inhibition.
