CJC-1295 with DAC is a 30-amino-acid synthetic analog of growth hormone-releasing hormone (GHRH) incorporating a Drug Affinity Complex (DAC)—a lysine-linked, maleimidopropionamide-derived moiety that enables covalent binding to serum albumin. This modification fundamentally distinguishes it from CJC-1295 without DAC (also catalogued separately in our catalog) and represents a different pharmacokinetic approach to sustained GHRH-receptor stimulation.

Like its non-DAC counterpart, CJC-1295 with DAC shares the same GHRH(1-29) core structure with four stabilizing amino acid substitutions (at positions 2, 8, 15, and 27) that resist enzymatic degradation. However, the DAC modification binds covalently to cysteine-34 on circulating serum albumin, creating an albumin-peptide complex that acts as a slow-release reservoir. This dramatically extends the peptide's half-life from approximately 30-60 minutes (without DAC) to approximately 6-8 days, allowing once-weekly dosing and sustaining GHRH-receptor stimulation and GH secretion over an extended period.

This sustained, non-pulsatile GH secretion pattern differs functionally from the pulsatile release mimicked by CJC-1295 without DAC and creates a distinct research profile: CJC-1295 with DAC is studied in models requiring sustained, continuous GH signaling, whereas the no-DAC variant is preferred for research examining physiological pulsatile GH dynamics or for combination protocols with GH secretagogues like ipamorelin.
