Glutathione (GSH, L-γ-glutamyl-L-cysteinyl-glycine) is a ubiquitous tripeptide found in all aerobic organisms and is the most abundant non-enzymatic antioxidant in mammalian cells, accounting for over 90% of non-protein cellular thiols. First characterized in the late 19th century, it remains one of the most extensively studied molecules in redox biology.
Glutathione's primary mechanism is maintaining cellular redox homeostasis. As a cofactor for glutathione peroxidase enzymes, it scavenges reactive oxygen and nitrogen species, converting toxic peroxides into non-toxic byproducts while itself being oxidized to GSSG, a reaction reversed by glutathione reductase using NADPH. Beyond this classical antioxidant role, more recent research has established glutathione as an active redox signaling molecule: through S-glutathionylation of proteins and shifts in cellular thiol redox state, GSH availability influences transcription factors including NRF2, PGC-1α, and p53, linking it to autophagy regulation, apoptosis, and mitochondrial homeostasis. Glutathione is also a direct nucleophile, forming conjugates with electrophilic compounds via glutathione-S-transferases as part of cellular detoxification pathways.
