These modifications dramatically reduce the peptide's binding affinity for IGF binding proteins (IGFBPs), which normally sequester native IGF-1 in circulation and limit its bioavailability within minutes of secretion.
The functional consequence is a research analog with a substantially extended half-life (approximately 20-30 hours, compared to 10-20 minutes for native IGF-1) while retaining full agonist activity at the IGF-1 receptor. Because IGFBP interference is minimized, IGF-1 LR3 is frequently used in laboratory settings as a tool for studying IGF-1 receptor pharmacology in isolation, including downstream Akt/mTOR signaling, cell proliferation, and protein synthesis pathways, without the confounding influence of binding protein sequestration seen with native IGF-1.
