Kisspeptin is a neuropeptide encoded by the KISS1 gene that activates a G-protein-coupled receptor known as KISS1R (formerly GPR54). It occupies the most upstream position in the hypothalamic-pituitary-gonadal (HPG) axis of any compound in our catalog: where hCG and HMG act directly on gonadal LH/FSH receptors, kisspeptin acts on GnRH neurons themselves, the neurons that initiate the entire reproductive hormonal cascade.
Kisspeptin/KISS1R signaling has been established through genetic knockout research as the most potent known stimulus for GnRH-induced gonadotropin release. KISS1R-deficient mice and humans with inactivating KISS1R mutations both fail to undergo normal pubertal development and exhibit hypogonadotropic hypogonadism, demonstrating the receptor's essential, non-redundant role in reproductive axis activation. Research using GnRH-neuron-specific receptor knockouts has further localized kisspeptin's primary site of action to the GnRH neuron itself, distinguishing its mechanism from gonadotropins that act peripherally on the gonads. Kisspeptin neurons have also been shown to express estrogen and progesterone receptors, linking this pathway to feedback regulation from peripheral sex steroids.
