KPV

KPV is a tripeptide composed of lysine, proline, and valine (Lys-Pro-Val), corresponding to the C-terminal fragment (residues 11-13) of alpha-melanocyte-stimulating hormone (α-MSH). Despite its minimal three-amino-acid structure, KPV retains a significant portion of the parent hormone's anti-inflammatory signaling while completely lacking α-MSH's pigmentation and appetite-related effects.

What makes KPV mechanistically distinctive is that it does not require melanocortin receptor binding to act. Instead, the peptide is actively transported into cells via the PepT1 di/tripeptide transporter, where it directly inhibits NF-κB, a central transcription factor controlling inflammatory gene expression, including the production of pro-inflammatory cytokines such as TNF-α, IL-1β, and IL-6. This receptor-independent mechanism distinguishes KPV from other melanocortin-derived peptides in our catalog, such as Melanotan 1 and Melanotan 2, which act through classical MC-receptor signaling. KPV is studied in laboratory settings primarily in the context of intestinal and epithelial inflammation models.
