SS-31 (elamipretide) is a synthetic four-amino-acid peptide (D-Arg-2'6'-Dmt-Lys-Phe-NH₂) belonging to the Szeto-Schiller class of mitochondria-targeted peptides. It carries a strong positive charge and an alternating cationic/aromatic side-chain motif that allows it to cross the plasma membrane in an energy-independent, non-saturable manner and accumulate at the inner mitochondrial membrane at concentrations 1,000 to 5,000-fold higher than in surrounding tissue.
SS-31's mechanism centers on selective binding to cardiolipin, an anionic phospholipid found almost exclusively on the inner mitochondrial membrane and essential for cristae structure and electron transport chain organization. By binding cardiolipin, SS-31 is proposed to stabilize membrane curvature, alter local electrostatics, and reduce mitotoxic interactions between cardiolipin and basic proteins, thereby supporting cristae integrity and ATP production efficiency. This places SS-31's research relevance in mitochondrial membrane biophysics, distinct from MOTS-C's gene-expression and AMPK-based mechanism elsewhere in our catalog, offering researchers two structurally and mechanistically different entry points into mitochondrial biology.
